Bossé Y, Maghni K, Hudson TJ. 1 ,25-Dihydroxy-vitamin D3 stimulation of bronchial smooth muscle cells induces autocrine, contractility, and remodeling processes. Physiol Genomics 29: 161–168, 2007. First published January 9, 2007; doi:10.1152/physiolgenomics.00134.2006.— Genetic variants in the vitamin D receptor (VDR) gene were recently associated with asthma. The biological mechanisms explaining this association are unknown but are likely to involve many cell types given the pleiotropic effect of its ligand, 1 ,25-dihydroxy-vitamin D3 [1 ,25(OH)2D3]. Considering the prominent role of bronchial smooth muscle cells (BSMCs) in the pathogenesis of asthma, experiments were conducted to explore the gene regulatory effects of 1 ,25(OH)2D3 in these cells. Using RT-PCR and Western blot, we showed that VDR is present both at the mRNA transcript and protein levels in human BSMCs. The functionality of the receptor was then demonstrated by showing a 200-fold change in the expression of the 24-hydroxylase (CYP24A1) gene following 1 ,25(OH)2D3 stimulation. Microarray experiments were then performed to identify differentially regulated genes and pathways in BMSCs treated or not with 1 ,25(OH)2D3. A total of 729 probe sets on the U133 plus 2.0 Affymetrix GeneChip showed foldchange differences above the 1.5 threshold using the Robust Multichip Average intensities. This corresponds to 231 unique genes that were upregulated and 215 unique genes that were down-regulated following 1 ,25(OH)2D3 stimulation. A high similarity between microarray and real-time PCR results was observed for 13 random genes, with a concordance correlation coefficient of 0.91. Real-time PCR was also performed to confirm the regulation of asthma candidate genes. To identify the biological relevance of this regulation, biological pathways analyses were performed. The most significant network of upregulated genes included genes involved in morphogenesis, cell growth, and survival as well as genes encoding structural proteins, which are potentially involved in airway remodeling.

متن کامل1 ,25-Dihydroxyvitamin D3 Induces Vascular Smooth Muscle Cell Migration via Activation of Phosphatidylinositol 3-Kinase
The steroid hormone 1 ,25-dihydroxyvitamin D3 [1 ,25-(OH)2D3] promotes vascular smooth muscle cell (VSMC) growth and calcification, but the precise mechanism by which 1 ,25-(OH)2D3 regulates VSMC migration is unknown. In rat aortic SMCs, we found that 1 ,25-(OH)2D3 (0.1 to 100 nmol/L) induced a dose-dependent increase in VSMC migration. This response required the activation of phosphatidylinosi...
متن کاملStimulation of calmodulin binding to skeletal muscle membrane proteins by 1,25-dihydroxy-vitamin D3.
Previous work has shown that 1,25-dihydroxy-vitamin D3 rapidly increases calmodulin levels of skeletal muscle membranes without altering the muscle cell calmodulin content. Therefore, the effects of the sterol on the binding of calmodulin to specific muscle membrane proteins were investigated. Soleus muscles from vitamin D-deficient chicks were treated in vitro for short intervals (5-15 min) wi...

Background and purpose : Toxoplasma gondii is an obligate interacelullar parasite that infect a wide variety of nucleated cells in its intermediate hosts. Tachyzoites of Toxoplasma gondii in able to multiply in mouse macrophages. Inducing synthesis of nitric oxide (NO) by macrophages is an important mechanism of the host defense against interacelullar infection in mice. The 1.25 dihydroxy vit...
متن کامل25-hydroxyvitamin D3-1alpha-hydroxylase is expressed in human vascular smooth muscle cells and is upregulated by parathyroid hormone and estrogenic compounds.
BACKGROUND 1,25(OH)2 vitamin D3 exerts multiple effects in human vascular smooth muscle cells (VSMCs). We therefore tested the possibility that VSMCs possess an endogenous 25-hydroxyvitamin D3-1alpha-hydroxylase system, the final enzyme in the biosynthetic pathway of 1,25(OH)2D3. METHODS AND RESULTS We assessed the expression and activity of 25-hydroxyvitamin D3-1alpha-hydroxylase by real-tim...
